SUMMARY:II Jornada de Igualdad de Biogipuzkoa: ¿dónde estamos? ¿qué tenemos por delante?
DESCRIPTION:Hace dos años\, presentamos el Plan de Igualdad 2024-2027 del Instituto. Al ser el ecuador del plan\, compartiremos con las compañeras y compañeros del Instituto la labor realizada estos dos años. Además\, compartiremos el análisis que hemos realizado en distintos ámbitos (trabajos académicos\, formación\, divulgación\, publicaciones\, proyectos y vida interna) sobre sesgo de género. Con toda esta información\, queremos plantear un diálogo abierto y sincero\, siguiendo los que ya hemos tenido en las mesad redondas de Emakumeak Zientzian\, para ser conscientes del trabajo que hay por delante en los dos siguientes años.

SUMMARY:Long-Read DNA and RNA Sequencing with Oxford Nanopore Technology (ONT): Comparative cDNA and Direct RNA Analysis for Lentiviral Vector Genome Integrity Assessment
DESCRIPTION:Oxford Nanopore long-read sequencing has transformed genomics and transcriptomics by enabling direct\, high-resolution analysis of RNA and DNA molecules\, including structural variants\, repetitive expansions\, and epigenetic modifications. In the NeuroRNA group\, we have applied this technology for several applications\, including the quality control (QC) of lentiviral vectors\, a critical step in gene therapy development. \nIn collaboration with VIVEbiotech and supported by the MedTech Initiative\, we developed a robust assay to assess the genome integrity of lentiviral vectors. Both cDNA and direct RNA (dRNA) sequencing were applied to purified viral RNA from concentrated preparations\, allowing comparison of sequencing efficiency\, coverage\, and fidelity across constructs of varying complexity. \nOur analysis highlights the strengths and limitations of cDNA versus dRNA sequencing for vector characterization. We also demonstrate that modifying construct orientation and structure reduces alternative splicing\, improving vector reliability. \n  \n\ntbd

SUMMARY:Development of lymphatic progenitors in vitro and their potential in biomedical applications
DESCRIPTION:The lymphatic system\, despite its widespread distribution and critical role in various biological processes\, remains relatively underappreciated. Recent studies have moved beyond viewing it merely as a system of conduits\, highlighting its significant involvement as a main driver of numerous diseases. However\, progress has been hindered by the difficulty in isolating primary lymphatic endothelial cells and the scarcity of reliable lymphatic progenitors derived in vitro. Consequently\, the development of functional in vitro models or advanced therapies for lymphatic and lymphatic-associated pathologies first requires establishing robust and adequate methods for generating lymphatic progenitors. This thesis addresses this need by exploring different strategies and tissue sources for obtaining in vitro differentiated lymphatic progenitors\, culminating in a comprehensive characterization of their potential in disease modelling\, cellular therapy\, and their translatability to human samples.

SUMMARY:Aproximación metabolómica en investigación clínica
DESCRIPTION:La Plataforma de Metabolómica forma parte de las plataformas de apoyo a la investigación del Instituto de Investigación Sanitaria Biobizkaia. \nLa Metabolómica comprende diversas técnicas de análisis (como la cromatografía y la espectrometría de masas) que permiten obtener el conjunto de metabolitos de un organismo (intermedios metabólicos o moléculas de bajo peso molecular) en un momento dado. La comparación de los metabolomas entre el grupo control y el grupo testado puede descubrir diferencias en sus perfiles\, pudiendo determinar qué metabolitos se asocian con dicha condición y podrían actuar como biomarcadores de diagnóstico o evolutivo de una enfermedad. \nLa Metabolómica tiene una aplicación crucial en los siguientes campos: \n\nBúsqueda de biomarcadores de diagnóstico y evolución en caso de enfermedad.\nEstudio de las perturbaciones metabólicas que se producen por la modificación o ausencia de un determinado enzima o proteína.\nEstudio de los lípidos a través de la Lipidómica.\nEstudios farmacéuticos.

SUMMARY:From intrinsic programs to tumor–microenvironment interactions in aggressive prostate cancer
DESCRIPTION:Understanding the drivers of aggressive prostate cancer remains a major clinical challenge due to treatment-related confounding and disease heterogeneity. A key objective is to identify molecular programs associated with metastatic potential and how they interact with the tumor microenvironment. \nWe use de novo metastatic disease as a model to study progression in the absence of therapeutic pressure. By integrating bulk and single-cell transcriptomics\, we have identified intrinsic transcriptional programs linked to metastatic potential in a treatment-naïve context. \nIn parallel\, we investigate how alterations enriched in advanced disease\, including PTEN loss\, shape tumor behavior within its tissue context. Our analyses indicate that PTEN loss is associated with remodeling of the tumor microenvironment\, including tumor–stroma communication\, with potential implications for disease progression. \nOur work\, carried out in close collaboration with clinical teams and patient-derived cohorts\, aims to understand how tumor programs\, genetic alterations\, and tissue context interact to drive aggressiveness within an evolutionary framework.
