SUMMARY:Understanding the role of Oncostatin M signalling in the remodelling of the breast cancer immune microenvironment
DESCRIPTION:Chronic inflammation is considered one of the main drivers of cancer progression and cytokines are key regulators of this inflammation. Recent data from our group support that cytokine Oncostatin M (OSM) acts as a key player in the interactions in the breast tumour microenvironment\, where OSM activates an intriguing pro-tumoral signalling in cancer-associated fibroblasts (CAFs) and cancer cells. Our recent unpublished work characterised the effects of OSM in immune remodelling and immune suppression. Data from our laboratory supports the idea that OSM\, mainly produced by myeloid cells\, has a potential immunosuppressive effect by increasing macrophage and neutrophil recruitment and favoring the secretion of cytokines related to protumoral inflammation. Our next steps are focused on investigating which are the specific mechanisms by which OSM-OSMR axis changes immune cell populations and their phenotype in the tumor microenvironment and decipher if this axis could be a promising target for the development of new cancer immunotherapies.

SUMMARY:Programa TREBEA: Desarrollo\, implementación y resultados preliminares / Evaluation of RNA-targeting CRISPR-Cas system for splicing modulation in Spinal Muscular Atrophy (SMA)
DESCRIPTION:Programa TREBEA: Desarrollo\, implementación y resultados preliminares  \nEl programa piloto TREBEA\, es un novedoso tratamiento integral para pacientes con Trastorno Límite de Personalidad que ha sido desarrollado con un enfoque interdisciplinar y transcultural en la Red de Salud Mental de Gipuzkoa y está siendo evaluado. Los pacientes son derivados desde 13 centros de salud mental de Gipuzkoa.  El programa tiene una duración de 16 semanas y está basada en la Terapia Dialéctica Conductual. La evaluación incluye una entrevista clínica estructurada (DIB-R)\, mediante la que se obtiene información relativa a los antecedentes\, curso clínico\, síntomas actuales\, comorbilidades\, tratamiento farmacológico\, funcionamiento psicosocial y otros factores significativos. Por otra parte\, se aplican diferentes cuestionarios de autoinforme como\, CORE-OM\, BSL-23 e ICG-TLP. Esta evaluación se realiza pre y post intervención.  Los resultados obtenidos hasta la fecha muestran una reducción significativa en la frecuencia de conductas autolesivas e impulsivas\, una disminución en la gravedad de los síntomas y una mejora en el funcionamiento psicosocial. Además\, hay reducción en el número de atenciones en urgencias y en los ingresos hospitalarios\, al concluir y tras 6 meses una vez finalizado el programa. \n  \nEvaluation of RNA-targeting CRISPR-Cas system for splicing modulation in Spinal Muscular Atrophy (SMA) \nManipulation of RNA-processing is a validated therapeutic strategy for multiple diseases. Synthetic antisense oligonucleotides (ASOs) are being used to generate alternative RNA isoforms that alleviate disease. Despite their progress\, they pose several challenges regarding ASO delivery and stability\, which limits their translation into therapies. CRISPR-dCas13 system represents an interesting alternative to synthetic ASOs for splicing manipulation. This system is based on catalytically inactive RNA-targeting Cas13 molecules\, that led by complementary guide-RNAs (gRNA) can bind the target RNAs\, modulating different processes as alternative splicing. This system offers advantages such as a low cost associated with its design\, the possibility of expressing different gRNAs simultaneously\, and the possibility of using viral vectors for its administration\, reducing treatment to a single dose. In this work\, we employ the CRISPR-dCas13 system to manipulate SMN2 splicing as a therapeutic approach for Spinal Muscular Atrophy (SMA). Our results show that CRISPR-dCas13 system can be used at several levels\, from target sequence identification in established cell lines to mRNA splicing modulation in patient-derived cellular models.
