SUMMARY:Las mil caras de la salud mental en la cuarta edición del ciclo “Zientzia – Historia – Ciencia”
DESCRIPTION:Con el lema “Las mil caras de la salud mental”\, se quiere dialogar con la ciudadanía sobre cómo ha ido cambiando la mirada sobre la salud mental y en cómo se ha gestionado en distintas épocas\, intentando desestigmatizarla. Para ello\, se realizarán varias actividades en el Museo Romano Oiasso de Irún del 19 al 22 de octubre. \nPara comenzar\, se realizarán tres charlas dialogadas para abordar distintos aspectos de la salud mental\, desde la perspectiva histórica y científica. \n\nEl lunes 19 de octubre\, se tratará sobre Salud Mental y Mujer con Cristina Ubani y Rafael Huertas.\nEl martes 20\, Enric Novella y Álvaro Muzquiz dialogarán sobre la historia de la psiquiatría y cómo se han tratado los malestares y los traumas.\nEl miércoles 21 se hablará de suicidio con Cristina Blanco y Alex Muela.\n\nLas sesiones serán moderadas\, entre otras\, por Itziar Vergara o Marialui Garcia. Todos los diálogos serán de 19:00 a 20:00\, estarán abiertos a la participación de las personas que acudan y la entrada será gratuita hasta completar aforo. \n\nEl jueves 22 de octubre se realizará la grabación en directo de un podcast de la mano de la asociación “Why not?”\, a partir de las 17:00.

SUMMARY:Characterization of skeletal muscle in a mouse model of ALS upon FOXO1 inhibition / Role of inflammation in lung cancer: MyD88 as the key player
DESCRIPTION:Amyotrophic lateral sclerosis (ALS) is a devastating neurodegenerative disease characterized by the loss of motor neurons and skeletal muscle wasting. Although growing evidence points to skeletal muscle as an active player in ALS\, its contribution to disease progression and its potential as a therapeutic target remain incompletely understood. We previously showed that inhibiting FOXO1\, a transcription factor involved in muscle atrophy and metabolism\, has beneficial effects in cellular and Drosophila models of ALS. In this study\, we investigated whether pharmacological FOXO1 inhibition could improve disease outcomes in the hSOD1G93A mouse model of ALS. Mice were treated with the FOXO1 inhibitor\, starting either before the onset of symptoms or after disease onset. We evaluated motor function\, survival\, body weight\, skeletal muscle pathology and muscle gene expression. Our findings revealed limited benefits of FOXO1 inhibition\, highlighting the complexity of skeletal muscle alterations in ALS and the need to explore complementary therapeutic strategies. \n\nNon-small cell lung cancer (NSCLC) often occurs with a well-defined inflammatory tumor microenvironment (TME). Continuous inflammatory conditions in the TME induce cell damage and death\, resulting in the release of damage-associated molecular patterns (DAMPs). DAMPs are sensed by toll-like receptors (TLR) and interleukin-1 receptor (IL-1R) families signaling through the adaptor protein MyD88. Many attempts have been made to modulate the effects of the receptors in cancer but being the central hub of these inflammatory cascades\, MyD88 modulation may be a more effective strategy. However\, the implication of MyD88 in coordinating cancer-intrinsic inflammation and cancer progression remains to be clearly explained. We are convinced that MyD88 may exert divergent effects depending on the oncogenic context\, but enhancing tumor progression in highly inflamed tumors. Our objective is to delve deeper into the multifaceted role played by MyD88 in NSCLC\, a highly prevalent and deadly disease\, to identify the specific contexts in which modulating MyD88 or its effectors may be an effective anti-tumor immunotherapy for the disease.

SUMMARY:Understanding repeat expansion diseases using population-scale genomic data
DESCRIPTION:DNA repeat expansions are a major cause of neurological disease\, but their study has historically been limited to clinically ascertained cohorts. My research has helped shift this perspective by combining genomic technologies with population-scale datasets to investigate repeat expansion diseases beyond affected individuals. We developed a clinical-grade pipeline for detecting pathogenic expansions using short-read genome sequencing\, enabling diagnoses in previously unsolved cases and supporting its adoption as diagnostic test in the National Health Service in the UK. Applying these approaches to over 500\,000 individuals revealed that pathogenic expansions are substantially more frequent than previously recognised\, suggesting disease prevalence may be two- to threefold higher than clinical estimates and challenging assumptions about penetrance and disease recognition\,  highlighting the need to identify modifiers of disease expression. Lastly\, analysing over 10\,000 family trios\, we quantified germline repeat instability and identified determinants of repeat-length mutations\, including risks associated with intermediate alleles. Together\, these advances establish a population-scale framework for understanding disease prevalence\, penetrance and repeat mutability\, informing genetic risk prediction and disease classification.
